Relative levels of cytokine mRNA transcripts in goat PBMCs pretreated with negative or TMEM63A siRNA. Goat PBMCs pretreated with TMEM63A siRNA were stimulated with PBS or rHco-gal-m. After the stimulation, changes in cytokine transcript levels in cells were detected by real-time PCR. Each stimuli is shown in one of the two graphs. (A) IL-10; (B) IFN-γ; (C) TGF-β1. Data are presented as the means ± SD; n = 3. Significant differences between the two stimulation conditions for each RNAi group are indicated by an asterisk (*, p < 0.001). A number sign (#) designates that the mean of the TMEM63A knockdown group differs significantly from the mean of the negative siRNA group (p < 0.001) for a stimulation condition. Representative data from one independent experiment with technical triplicates are shown that is representative of three independent experiments.
