Cryptosporidium parvum inhibited autophagy of HCT-8 cell by regulating miR-26a and miR-30a expression. MiR-26a-mimic, miR-26a-inhibitor, miR-30a-mimic, miR-30a-inhibitor or ssRNA (negative control) was transfected into HCT-8 cells, and the cells were infected with C. parvum sporozoites at 24 h post-transfection. The protein samples were collected after 12 h infection and analyzed by western blotting with rabbit anti-LC3, anti-Beclin1, anti-p62 and anti-GAPDH antibodies (A, E). The results were further analyzed by grayscale analysis from three independent experiments (B–D, F–H). miR-26a-mimic, miR-30a-mimic or ssRNA (negative control) was transfected into HCT-8 cells, and the cells were infected with C. parvum sporozoites at 24 h post-transfection. The expression of miR-26a and miR-30a was detected using qPCR. A one-way ANOVA assay with Tukey-Kramer post hoc test was used for analyzing data. Data are expressed as the mean ± SD from three independent experiments (*P < 0.05, **P < 0.01, ns = no significant differences)
