PCR-RFLP assay for polymorphism in laboratory P. papatasi . Template DNA (as indicated for each lane) was amplified by PCR then subjected to digestion with Alu I and electrophoretically separated on a 4% Metaphor agarose gel. Lane: Std, DNA size standards; 1, no template negative control; 2, wild type PpAChE1 plasmid; 3, PpAChE1-G119S plasmid; 4–6, genomic DNA extracted from individual P. papatasi colony females fed sugar water only, no blood.
