Real-time PCR analysis after transfection of siRNA. Acanthamoeba castellanii were transfected with siRNA designed against ACA1_114460, ACA1_091500, and ACA1_362260 prior to bacterial ingestion for 12 h. Knockdown of gene expression was confirmed by real-time PCR (a ACA1_114460, b ACA1_091500, and c ACA1_362260). Data are expressed as mean ± SD of three separate experiments. Asterisks denote statistically significant differences (** P < 0.01, *** P < 0.001, and **** P < 0.0001) between the control group and the siRNA-transfected group
